SNAP-8 (Acetyl Octapeptide-3) is a synthetic peptide that mimics the N-terminal structure of the SNAP-25 protein, developed by the internationally renowned cosmetic ingredient supplier Lipotec (now part of Lubrizol) . It is an elongation of the popular Acetyl Hexapeptide-3 (ARGIRELINE®) by two amino acid residues, designed to achieve better anti-wrinkle effects . SNAP-8 competes with SNAP-25 for a position in the SNARE complex, thereby modulating its formation, reducing neurotransmitter release, and achieving muscle relaxation and expression line fading .
Key Scientific Value & Findings
Unique Mechanism of Action - Botox-like but Non-Paralytic: SNAP-8 has a similar target to Botox (both target SNAP-25 protein), but with different mechanisms of action :
Botox: Cleaves the SNAP-25 protein, causing neuromuscular junction paralysis - potent but irreversible
SNAP-8: As a SNAP-25 mimetic peptide, it competitively occupies positions in the SNARE complex, modulating complex formation and relaxing muscles rather than paralyzing them, thus offering higher safety
Superior to Acetyl Hexapeptide-3: SNAP-8 was designed by adding two amino acids to Acetyl Hexapeptide-3 to achieve better wrinkle improvement effects . Studies indicate that SNAP-8 is more effective than Acetyl Hexapeptide-3 .
SNARE Complex Modulation: SNAP-8 is a mimic of the N-terminal end of SNAP-25 which competes with SNAP-25 for a position in the SNARE complex, thereby modulating its formation . The SNARE complex plays a key role in neurotransmitter release; modulating its formation regulates the transmission of muscle contraction signals.
High Safety Profile: Compared to Botox, SNAP-8 offers higher safety . Botox cleaves the SNAP-25 protein, while SNAP-8 merely competitively occupies positions without destroying protein structure, resulting in mild and reversible effects.
